Summary |
We performed single-nucleus RNA-seq using 10X on 84 randomly sampled participants from the METabolic Syndrome In Men (METSIM) cohort. The purpose of the study was to characterize cell type composition changes associated with metabolic diseases such as obesity and type 2 diabetes.

Overall design |
Single cell RNA-seq samples were multiplexed to increase sample size. Fresh-frozen subcutaneous adipose tissue biopsies from 4 donors were pooled immediately prior to isolation. We then isolated nuclei using mincing, washing, and straining. Single nucleus sequencing libraries were prepared using the 10X single cell 3' v3.1 kit and the 10X Chromium platform. The resulting libraries were sequenced on an Illumina NovaSeq targeting 200 million reads per sample. Samples were demultiplexed with Demuxlet.
