Treatment protocol |
Cells were incubated in the presence or absence of Pioglitazone for 6 hours prior to RNA harvesting.

Growth protocol |
Freshly isolated PBMCs were cultured in RPMI/10% FBS in the presence or absence of 1µM pioglitazone (Sigma-Aldrich, St. Louis, MO, USA) for various timepoints. For microarray studies, PBMCs were cultured with pioglitazone for 6 h, harvested and RNA isolated.

Extraction protocol |
Total RNA was isolated with TriPure (Roche). For microarray analysis, RNA was further purified and concentrated using an RNeasy kit (Qiagen).

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from total RNA (Expression Analysis Technical Manual, Affymetrix).

Biotin-labeled cDNA was fragmented and hybridized to Affymetrix Human U133 Plus 2.0 GeneChip arrays (Santa Clara, CA) according to the manufacturer's protocol.

GeneChip arrays were scanned by GeneChip Scanner 3000 7G according to the Affymetrix Expression Analysis Technical Manual.
