1mm x 1mm biopsy tissues were placed under an Olympus model SZX12 stereomicroscope using 90x magnification. The biopsy tissue was manually microdissected at 4°C in RNALater for glomerular and tubular compartments using fine tip forceps. The corresponding tubulointerstitial and vascular compartment was placed into RLT solution. For easier designation we called this component tubules. Dissected tissue was homogenized using a Powergen125 (Fisher) homogenizer and stored at -80°C. RNA was prepared using RNAeasy mini columns (Qiagen) according to manufacturer's instructions. RNA quality and quantity was determined using a Lab-on-Chip Total RNA PicoKit (Agilent BioAnalyzer). Only samples without evidence of degradation were used.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Drosophila Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
The data were scanned using a GeneChip scanner in accordance with the Affymetrix protocol.

Data processing |
Data were normalized in GeneSpring software using the RMA16 summarization algorithm. Baseline is the median of all samples.
