Biotinylated cRNA was prepared from 100 ng total RNA according to the standard Affymetrix protocol (WT Expression Kit User Manual, 2010, Affymetrix).

Following fragmentation, 10 ug of cRNA was hybridized for 20 hr on Human Gene 2.1 ST arrays incubated at 48C. GeneChips were washed and stained using the Gene Atlas Fluidics Station.

HuGene 2.1 ST arrays were scanned using the Gene Atlas Imaging Station.

Data processing |
Data was analyzed with R/Bioconductor software and normalized using RMA. An ANOVA was used to identify differentially expressed genes. An FDR of 0.05 was used to account for multiple testing.
