Samples mds001-004, 013, 019 and 020 were derived from in vitro myeloid suppressors in IL-6 and GM-CSF according to Marigo et. Immunity 32: 790 (2010). Samples mds010 and 011 are spleen myeloid suppressors where Gr-1+ cells are depleted from the postively-sleceted CD11b+ fraction. The spleen samples originate from mice bearing EL4 flank tumors. Samples 021-023 CD14+ HLA-DR-/low were isolated from freshly obtained peripheral blood with consent from Head & Neck Squamous Cell Carcinoma patients undergoing surgical treatments at Johns Hopkins Hospital and Johns Hopkins Bayview Hospital

Following fragmentation, 5-6 micrograms of biotin-labeled targets were hybridized for 16 hr at 45C on an array using the Affymetrix GeneTitan system. Following hybridization, HT_MG-430_PM and HT_HG-U133_Plus_PM plate arrays were washed and stained in the Affymetrix GeneTitan system.
