Growth protocol Samples mds001-004, 013, 019 and 020 were derived from in vitro myeloid suppressors in IL-6 and GM-CSF according to Marigo et. Immunity 32: 790 (2010). Samples mds010 and 011 are spleen myeloid suppressors where Gr-1+ cells are depleted from the postively-sleceted CD11b+ fraction. The spleen samples originate from mice bearing EL4 flank tumors. Samples 021-023 CD14+ HLA-DR-/low were isolated from freshly obtained peripheral blood with consent from Head & Neck Squamous Cell Carcinoma patients undergoing surgical treatments at Johns Hopkins Hospital and Johns Hopkins Bayview Hospital

Extraction protocol Total RNA was extracted using Trizol (Invitrogen) according to the manufacturer's protocol
Label biotin
Label protocol Biotinylated cRNA was prepared from 100 ng of toal RNA according to the standard Affymetrix 3’IVT Express protocol (GeneChip 3’ IVT Express Kit 702646 Rev. 8, 2008-2010, Affymetrix).
 
Hybridization protocol Following fragmentation, 5-6 micrograms of biotin-labeled targets were hybridized for 16 hr at 45C on an array using the Affymetrix GeneTitan system. Following hybridization, HT_MG-430_PM and HT_HG-U133_Plus_PM plate arrays were washed and stained in the Affymetrix GeneTitan system.
Scan protocol HT_MG-430_PM and HT_HG-U133_Plus_PM plate arrays were scanned using the Affymetrix GeneTitan system.

Data processing Probe signals were quantile normalized and summarized by the RMA algorithm using the Affymetrix Expression Console Software v1.1.
