Extraction protocol The blood was centrifuged at 1000 g for 10 min, and buffy-coat was frozen in aliquots at −80 °C. DNA was isolated from the buffy-coats using the salt precipitation method applying either a manual protocol or a semi-automated protocol based on the Autopure System (Qiagen, Hilden, Germany). Bisulphite treatment of 500 ng template genomic DNA was carried out with the EZ-96 DNA methylation kit (Zymo Research, Orange County, USA) following the manufacturer’s protocol.
