Treatment protocol |
Male rats were treated with 50mg PB per 1000g body weight (5-ethyl-5-phenylbarbituric acid sodium salt; Sigma, St. Louis, MO) by a single gavage or via drinking water for a period of 14 days. Female Wistar rats were treated with 100mg CPA per kg body weight (Cyproterone acetate; Sigma, St. Louis, MO) by a single gavage on one day or on 6 consecutive days. Oil-treated rats served as control.

Growth protocol |
Han-Wistar rats were obtained from Charles River (FRA). They were kept under standardized SPF-conditions. All experiments were approved by the “Committee of Animal Protection” of the Austrian government and performed according to Austrian regulations.

Cells were homogenized with Qiazol (Qiagen, Ger) and total RNA was subsequently extracted with the miRNeasy Mini Kit (Qiagen, Ger) according to the manufacturer's instructions. RNA quality was assessed by measuring the RIN (RNA Integrity Number) using an Agilent 2100 Bioanalyzer. RNA was stored at −80°C until GeneChip® experiment analysis.

250ng of total RNA were amplified and labeled using the Affymetrix 3` IVT Express Kit (Catalog Number 901229, Affymetrix, Santa Clara,CA)

Affymetrix GeneChip® Rat 230 2.0 arrays were washed, stained and scanned according to the protocol described in GeneChip® 3` IVT Express Labeling Assay User Manual (Fluidics Protocol FS450_0002)

Scanning was done on an Affymetrix GeneChip Scanner 3000 7G
|
Data processing |
The raw microarray data was normalized using the RMA method implemented in R/Bioconductor.
