| Treatment protocol |
The study was designed as a phase I randomized, double-blind, placebo-controlled crossover study of the pharmacodynamics, safety, and clinical efficacy of AMG 811 in patients with DLE. Patients received a single subcutaneous dose of AMG 811 (180 mg) or placebo. The patients in sequence 1 received AMG 811 followed by placebo, while those in sequence 2 received placebo followed by AMG 811.

Total RNA was isolated from PAXgene whole blood tubes (BD Cat # 762165) using the PAXgene RNA Kit. (Qiagen Cat # 762164) on the Qiacube automated sample prep system

| Scan protocol |
All arrays were scanned on the Agilent Surescaner Scanner (Agilent p/n G2600D). Agilent Scan Control Software version 9.1.5.1 was used for array scanning. Array files were processed using Agilent Feature Extraction software 11.0.1.1.

| Data processing |
The scanned images were analyzed using default parameters (protocol GE1-v1_91 and Grid: 026822_D_F_20100209) to obtain background subtracted and spatially detrended Processed Signal intensities. Features flagged in Feature Extraction as Feature Non-uniform outliers were excluded.
