Treatment protocol |
The right external jugular vein was connected to the common carotid artery by end-to-end anastomosis using a 10.0 silk suture (Ethicon Inc., Somerville, NJ).

Growth protocol |
Wistar rats (250-350g) were anesthetized with a mixture of ketamine (50 mg/kg) and xylazine (10mg/kg).

Arterialized vein segments were collected at 3 days (n=3) and 28 days (n=3) after surgery. Normal jugular veins (n=5) and carotid arteries (n=2) were used as controls.

Biotin-labelled cRNA was produced form 1 ug of total RNA. First and second-strand cDNA was synthesized according to manufacturer’s instruction. cRNA was obtained by in vitro transcription reaction.

Ten microgram of fragmented cRNA was incubated with slide array for 24h at 37 ºC, under 300 RPM shaking to ensure homogenization.

Slides were scanned using GenePix 4000B Array Scanner using Scan Codelink settings.

Data processing |
The raw data were normalized using CyclicLoess, the most effective method for CodeLink Bioarray data. Data was also filtered for good intensity of the signal and removing genes with a low-intensity signal in at least 50% of the samples of each group.
