Total RNA was isolated, treated with DNase I, and cleaned up using a Qiagen RNeasy MinElute Cleanup Kit. RNA concentrations were determined using NanoDrop (Thermo Fisher Scientific, Waltham, MA, USA) and RNA quality was evaluated using the Experion electrophoresis system (Bio-Rad Laboratories, Hercules, CA, USA).

Double-stranded cDNAs were synthesized from 1 μg of total RNA through reverse transcription with an oligo-dT primer containing the T7 RNA polymerase promoter and converted to double-strand using the cDNA Synthesis System (Invitrogen). Biotin-labeled cRNA was generated from the cDNA. All cDNA and cRNA target preparation steps were processed on a Caliper GeneChip Array Station from Affymetrix.

Biotin-labeled cRNA was generated from the cDNA and used to probe a Human Genome HT_HG-U133A plate (Affymetrix, Sunnyvale, CA, USA) Array hybridization, washing, and scanning were performed according to the manufacturer’s recommendations.
