Genomic DNA was extracted using the Blood and Cell Culture Maxi Kit or Mini Kit (Qiagen) Genomic DNA from naïve cells or cells surviving lethal H5N1 infection was extracted. The integrated sgRNA cassette was PCR amplified from genomic DNA. Illumina barcodes were incorporated in the PCR primers.
