Sample type |
RNA |
Source name |
Homograft B16F10L-luciferase tumor 7 days post sub cutaneous inoculation
Organism |
Mus musculus |
Characteristics |
gender: Male strain: C57BL/6J genotype: ApoA-I Tg
Treatment protocol |
Tumor tissue from one flank was resected on day 7 post inoculation and stored in RNA later.
Growth protocol |
Eight to ten (8-10) week male and female animals were inoculated subcutaneously on both flanks with 105 B16F10L-luciferase tumor cells per site (2 sites/flank).
Extracted molecule |
total RNA |
Extraction protocol |
Tumor tissue (30 mg) was homogenized with zircodium oxide beads using the Bullet blender tissue homogenizer (Next Advance, Troy, NY, USA) and total RNA was prepared (RNeasy Mini kit, Qiagen, Valencia, CA) with on column DNase treatment (Cat# 79254, Qiagen).
Label |
Biotin
Label protocol |
Total RNA was amplified and biotinylated for hybridization using Illumina TotalPrep RNA Amplification Kit (AMIL1791, Applied Biosystems, Life Technologies) according to the manufacturer’s protocol.
Hybridization protocol |
Standard Illumina hybridization protocol
Scan protocol |
Arrays were scanned in an Illumina Bead Station, and the images processed using Illumina Bead Studio software.
Data processing |
The data were normalised using quantile normalisation with IlluminaGUI in R
