Treatment protocol |
POWV-LI-9 was allowed to adsorb to ~80% confluent hBVP monolayers for 1 h. Following adsorption, monolayers were washed with PBS and grown in supplemented Pericyte Media with 10% FBS. RNA was extracted 1-3 dpi. Mock-infected cells were used as control.

Growth protocol |
hBVPs (passage 5), derived from elutriation of dispase-dissociated normal human brain cortex tissue were purchased from Cell Biologics (H-6023), and were grown in endothelial cell basal medium-2 MV (EBM-2 MV; Lonza) supplemented with EGM-2 MV SingleQuots (Lonza) and incubation at 37°C and 5% CO2.

Total RNA was purified using RNeasy (Qiagen) following the manufacturer protocol.

Fragmented and labeled single-stranded cDNA was generated from total RNA using the GeneChip WT PLUS Reagent Kit (Affymetrix).

Samples were hybridized to GeneChip Human Clariom S Arrays (Affymetrix) using the Hydridization, Wash and Stain kit (Affymetrix).

Scan protocol |
Array scanning was performed according to the manufacturer's instruction (Affymetrix). Arrays were scanned at the Stony Brook Genomics Core Facility.

Data processing |
Raw data was processed with the Expression Console software (Affymetrix) for background correction and normalization. Data analysis and statistical evaluations were performed with Transcriptome Analysis Console (TAC) Software (Affymetrix).
