Title |
Liver from the rats treated with 1.5 mg/kg animal of dexamethasone and 0.7 g/kg animal of AICAR for 8 hours, rep 1. |
Sample type |
RNA |
|
|
Source name |
Rat liver, excised 8 hours
|
Organism |
Rattus norvegicus |
Characteristics |
agent: dexamethasone + AICAR gender: male strain: Sprague-Dawley weight: 180-200g
|
Treatment protocol |
The animal study was approved by the NICHD Animal Care and Use Committee (protocol numbers: ASP07-016 and ASP09-030). Male Sprague-Dawley rats (180-200g) were injected with either AICAR (0.7 g/kg animal) or physiologic saline into their peritoneal cavities. Animals in each group were also injected intramuscularly with dexamethasone (1.5 mg/kg) or normal saline. Eight hours after the injection, animals were euthanized using CO2, and their livers were harvested for total RNA purification.

Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted by using RNAeasy mini kit (Qiagen, Valencia, CA)
|
Label |
biotin
|
Label protocol |
cRNA was produced and labeled with the One-Cycle Target Labeling and Control Reagents Kit (Affymetrix, Inc, Santa Clara, CA).

|
|
Hybridization protocol |
We employed a standard hybridization method indicated by Affimetrix.
|
Scan protocol |
We used the GeneChip Fluidics Station 450 (Affimetrix) and the GeneChip Scanner 3000 7G System for washing and scanning the micorarrays.
