Title |
Enriched |
Sample type |
genomic |
Source name |
enriched HMEC-hTERT-p53DD-p110a(H1047R)
Organism |
Homo sapiens |
Characteristics |
cell type: BEZ235-selected immortal human mammary epithelial cells containing randomly mutagenized PIK3CA(H1047R)
Growth protocol |
HMECs were grown according to standard conditions.
Extracted molecule |
genomic DNA |
Extraction protocol |
Genomic DNA was extracted from HMEC cultures using Qiagen DNeasy mini Kit. DNA quality and quantity was assessed using Nanodrop, and agarose gel electrophoresis.
Label |
biotin
Label protocol |
As per manufacturer (Affymetrix)
Hybridization protocol |
DNA was restriction digested, PCR amplified, fragmented, labeled and hybridized to each array according to the manufacturer's instructions.
Scan protocol |
The Arrays were then washed using Affymetrix fluidics stations, and scanned using the Gene Chip Scanner M10.
Description |
BEZ235-selected immortal human mammary epithelial cells containing randomly mutagenized PIK3CA(H1047R)
Data processing |
CEL files and CHP files (generated by Affymetrix GTYPE using the DM algorithm) were processed by dChip and model-based expression performed using the PM/MM model and copy number calls generated as compared to Affymetrix normal controls.
