Treatment protocol Cell pellets were pretreated with 1ul Zymolyase for 1 hour at 30C prior to plasmid isolation
Growth protocol Strains used in the study include BC187, BY4743, NCYC3290, and YPS128. Strains were grown in YPD medium containing 0.7M NaCl and G418 (200mg/L) for plasmid selection, in flasks at 30°C. Frozen library-transformed stocks were thawed and placed in 100ml of liquid YPD with 0.7M NaCl and G418 (200mg/L) at a starting OD600 of 0.05. The pooled growing cultures were transferred to fresh media after 5 generations to keep cells in log phase. Cells were harvested after 10 generations and cell pellets were stored at -80°C.
Extracted molecule genomic DNA
Extraction protocol Plasmids were isolated from cell pellets using the Qiagen Spin MiniPrep Kit.
Libraries were constructed using PCR where plasmid barcodes were amplified with multiplex primers containing Illumina adapters and unique 10bp index tags to allow for sample identification after sequencing, as described in Magtanong et al., 2011 and Piotrowski et al., 2015.
