Magnaporthe appressoria: wt vs. bip1 deletion mutant

(Submitter supplied) The identification of genes regulated by the transcription factor Bip1 during appressorial development. The rice blast fungus Magnaporthe grisea develops melanized appressoria with high turgor pressure that inject the fungus into epidermal host cells. A screen for M. grisea non-pathogenic mutants recovered mutant M763 with reduced penetration efficiency and infectious growth. Complementation analysis identified a gene inactivated in M763 that encodes a novel basic leucine zipper (bZIP) transcription factor named Bip1 (B-ZIP Involved in Pathogenesis1). more...

Digital gene expression tag profiling of M. grisea G11 and T146

(Submitter supplied) The rice blast fungus Magnaporthe oryzae is a major constraint to the world food security. To study the intracellular mechanism involved in its pathogenic morphogenesis, digital gene expression tag profiling was used to compare a previously generated temperature sensitive mutant, T146, which stopped pathogenic morphogenesis at 33°C, with its related wide type, Guy11. 416 genes were detected as differential expressed genes, 26 of which were specifically expressed in Guy11 but stopped expression in the mutant. more...

Transcriptome profiling of the rice blast fungus during invasive plant infection and in vitro stresses

(Submitter supplied) Rice blast is the most threatening disease to cultivated rice worldwide. Magnaporthe oryzae, its causal agent is likely to encounter environmental challenges during invasive growth in the host plants that require shifts in gene expression to establish a compatible interaction. Here, we tested the hypothesis that M. oryzae have similar gene expression patterns in planta, versus during in vitro stresses. more...

Ca2+/calcineurin/CRZ1 dependent gene expression in Magnaporthe oryzae

(Submitter supplied) The Ca2+/calcineurin signaling pathway is a central conduit regulating growth, development, and virulence of fungal pathogens infecting plants and human. We have analyzed global gene expression profiles during Ca2+ treatment in the rice blast fungus, Magnaporthe oryzae. An immunosuppressive drug, FK506, and the knock-out mutant of a transcription factor, MoCRZ1, were included to analyze calcineurin- and/or CRZ1-dependent gene expression, respectively. more...

ChIP-chip of MoCRZ1

(Submitter supplied) CRZ1 (Calcineurin Responsive Zinc finger 1) is a conserved transcription factor that relays Ca2+/calcineurin signals within the cell. To catalog genome wide locations that MoCRZ1 binds to, we have conducted chromatin immunoprecipitation coupled with microarray (ChIP-chip) in the rice blast fungus, Magnaporthe oryzae. Fungal mycelia were treated with 200mM CaCl2 to activate translocation of MoCRZ1 to the nucleus, thereby binding to the promoter regions of downstream genes, and CaCl2 together with 10ug/ml FK506 to block these phenomena. more...

Magnaporthe oryzae Gene Expression During Biotrophic Invasion of Rice (Agilent Array, G4137B)

(Submitter supplied) The hemibiotrophic fungus Magnaporthe oryzae produces specialized biotrophic invasive hyphae (IH) that alter membrane structure and defense responses in invaded rice cells. IH successively invade live neighbor cells, apparently through plasmodesmata. Understanding fungal and rice genes that contribute to biotrophic invasion has been a challenge because so few plant cells have encountered IH at the earliest infection stages. more...

Differential gene expression of Appresorium vs Vegetative mycelium of Magnaporthe using oligoarray

(Submitter supplied) Rice blast, caused by the fungal pathogen Magnaporthe grisea, is a devastating disease causing tremendous yield loss in rice production. The public availability of the complete genome sequence of M. grisea provides ample opportunities to understand the molecular mechanism of its pathogenesis on rice plants at the transcriptome level. To identify all the expressed genes encoded in the fungal genome, we have analyzed the mycelium and appressorium transcriptomes using MPSS, RL-SAGE and oligoarray methods. more...
