Cell-free lncRNA expression signatures in urine serve as novel noninvasive biomarkers for diagnosis and recurrence prediction of bladder cancer

(Submitter supplied) To identify lncRNAs dysregulated in BC, we assessed the lncRNA expression profile in pooled urine samples of 10 BC patients and 10 controls by microarray. A total of 3093 lncRNAs determined differential expression (fold change>=2.0) between BC patients and controls. Among them, 1680 lncRNAs were up-regulated and 1413 lncRNAs were down-regulated in BC. Of all the differentially expressed lncRNAs indicated by lncRNA expression profiling, 26 lncRNAs (fold change>25) were firstly selected for further evaluation, in which 16 lncRNAs were up-regulated and 10 lncRNAs were down-regulated in BC.

Genome-wide DNA methylation study in bladder cancer

(Submitter supplied) Genome-wide DNA methylation profiles were determined on a set of fresh 44 bladder cancer tissues using normal blood as control. DNA amplicons were prepared using Differential Methylation Hybridization (DMH) method, subsequently hybridized on to the Agilent Human CpG island Microarray. Gene expression signatures in a human bladder cancer cell line SW780 transfected with miR-137 precursor molecule

(Submitter supplied) To clarify the effect of miRNAs, we carried out a gene expression microarray analysis of SW780 cells transfected with a miR-137 precursor or a negative control. We found that 1,326 probe sets (1,016 unique genes) were downregulated (>2-fold) by ectopic miR-137 expression, including the previously reported miR-137 target genes CDK6, CDC42 and AURKA. Moreover, Gene Ontology analysis revealed that genes related to the cell cycle were significantly enriched among the affected genes.

Genome-wide DNA methylation analysis of non-cancerous urothelium obtained from patients with urothelial carcinomas and corresponding cancerous tissue

(Submitter supplied) The aim of this study was to establish criteria for carcinogenetic risk estimation in the urothelium. Forty-six samples of non-cancerous urothelium showing no remarkable histological findings obtained from patients with urothelial carcinomas (N), and 46 samples of the corresponding cancerous tissue (T), i.e. 92 tissue samples in total, were analyzed. Genome-wide DNA methylation analysis was performed using the Infinium HumanMethylation 450K BeadChip.
