Functional investigation of miRNAs by characterization of SH-SY5Y cells overexpressing wild type or mutant miRNA genes

(Submitter supplied) Genome wide mRNA and miRNA profiling was performed in SH-SY5Y cells stably overexpressing wild type or mutant MIR204 or MIR618. Mutants came from a large scale genetic screening of brain expressed miRNA genes in patients with schizophrenia or idiopathic generalized epilepsy and in control individuals. Based on enrichment of the variants with the schizophrenic or epileptic phenotype and based on impact prediction, two variants, one near MIR204 (rs7861254) and one in MIR618 (rs2682818) were selected for functional validation. more...

Dynamic time course miRNA profiling of human skeletal muscle cell differentiation.

(Submitter supplied) The purpose of this study was to determine the miRNA expression profile of in vitro differentiation of human skeletal muscle cells and to couple changes in individual miRNA expression to transcriptional output of target genes. miRNA expression profiling at six different time points during the in vitro differentiation process of human skeletal muscle cells from six subjects. RNA was harvested from myoblasts before induction of differentiation and at every other day for 10 following days.

Deregulated MicroRNAs in Cancer-Associated Fibroblasts from Front Tumor Tissues of Lung Adenocarcinoma as Potential Predictors of Tumor Promotion

(Submitter supplied) Cancer-associated fibroblasts (CAFs) promote tumor progression through several mechanisms. MicroRNAs (miRNAs) play a key role in CAFs tumor-promoting properties, however, their role in CAFs from different topological areas in lung cancer progression remains unclear. This study aimed to characterize the miRNA expression profile of fibroblasts isolated from matched tumor front (F-CAFs), inner tumor (In-CAFs), and normal adjacent tissue (NFs) in lung adenocarcinoma, using microarray analysis and RT-qPCR. more...

MicroRNA expression profiling of NGF-treated PC12 cells revealed a critical role for miR-221 in neuronal differentiation

(Submitter supplied) MicroRNAs (miRNAs) are small non-coding RNAs that control protein expression through translational inhibition or mRNA degradation. MiRNAs have been implicated in diverse biological processes such as development, proliferation, apoptosis and differentiation. Upon treatment with nerve growth factor (NGF), rat pheochromocytoma PC12 cells elicit neurite outgrowth and differentiae into neuron-like cells. more...

Distinct cognitive effects and underlying transcriptome changes upon inhibition of individual miRNAs in hippocampal neurons

(Submitter supplied) MicroRNAs (miRNA) are small, non-coding RNAs mediating post-transcriptional regulation of gene expression. miRNAs have recently been implicated in hippocampus-dependent functions such as learning and memory, although the roles of individual miRNAs in these processes remain largely unknown. Here, we achieved stable inhibition using AAV-delivered miRNA sponges of individual, highly expressed and brain-enriched miRNAs; miR-124, miR-9 and miR-34, in hippocampal neurons. more...

High-throughput sequencing identifies miRNAs and their targets involved in porcine muscle development

(Submitter supplied) To identify miRNAs involved in muscle development, three independent small RNA libraries from porcine skeletal muscle at the age of 63d、98d and 161d were constructed. A total of 4105184, 4606375 and 5113984 mappable small RNA sequences were generated in the three small RNA libraries, respectively, and 887 known and 472 new candidate miRNAs were obtained.

MicroRNAs Expression Profiles Differ between Primary Myofiber of Lean and Obese Pigs breeds

(Submitter supplied) Purpose: MicroRNAs (miRNAs) are non-coding small miRNA ~22 nucleotides in length and play a vital role in muscle development by binding to messenger RNAs (mRNAs). Large White (LW, a lean type pig) and Meishan pigs (MS, a Chinese indigenous obese breed) have significant postnatal phenotype differences in growth rate, muscle mass and meat quality, and theses difference are programmed during prenatal muscle development. To shed directly light on the miRNA transcriptome difference in prenatal muscles between these two distinct pig breeds. more...
