Transcriptome profile of liver at different physiological stages in laying hens

(Submitter supplied) In this study, RNA-Seq technology was adopted to investigate the differences in expression profiles of the hepatic lipid metabolism-related genes and the associated pathways between juvenile and laying hens. RNA-Seq analysis was carried out to estimate total RNA harvested from the liver of juvenile hens (n = 3) and laying hens (n = 3). Compared with juvenile hens, 2574 differentially expressed (DE) genes (1487 down and 1087 up) with P ≤ 0.05 were obtained, and 955 of these genes were significantly DE (SDE) at a false discovery rate (FDR) of 0.05 and fold-change ≥ 2 in laying hens. more...

miRNA expression profiling in liver of juvenile and egg-laying hens

(Submitter supplied) A total of 565 miRNAs annotated in miRBase 20.0 were identified to be expressed in the liver of hen by high-throughput sequencing three biological reduplication libraries in juvenile and egg-laying hens, respectively. Compared with juvenile hen, 80 miRNAs (67 down-regulated and 13 up-regulated) were verified to be significant differential expression (SDE) in egg-laying physiological stage. Among these, miR-22-3p has the highest abundant expression, and miR-146b-5p has the highest fold-change. more...

RNA-seq analysis of differential gene expression in liver from lactating dairy cows divergent in negative energy balance

(Submitter supplied) Background: The liver is central to most economically important metabolic processes in cattle. However, the changes in expression of genes that drive these processes remain incompletely characterised. RNA-seq is the new gold standard for whole transcriptome analysis but so far there are no reports of its application to analysis of differential gene expression in cattle liver. We used RNA-seq to study differences in expression profiles of hepatic genes and their associated pathways in individual cattle in either mild negative energy balance (MNEB) or severe negative energy balance (SNEB). more...

Hepatic lipid metabolism in chicken lines divergently selected for abdominal fat content

(Submitter supplied) We performed gene expression profiling on chicken livers at five important embryonic developmental stages (E7, E12, E14, E17 and E21), which were selected from chicken lines with significant differences in abdominal fat content. Each developmental stage had its unique gene expression pattern and stage-specific differentially expressed genes (DEGs). Furthermore, the three rapid growth periods (E12, E14 and E17) had similar expression patterns. more...

Transcriptome profiling of feeding-to-fasting transition in chicken liver using a chicken 20K oligo microarray

(Submitter supplied) Four weeks old male broilers, fed ad libitum or fasted for 16h or 48h were used to describe the evolution of global gene expression profiles in chicken liver during a 48h fasting period using a chicken 20K oligo array. Among the 20460 oligos on the microarray, 13057 were identified as aligning with a unique coding region of the 2.1 Washington University assembly of the chicken sequence genome. So the statistical analyses were performed on this 13057 gene set. more...

Identification of key genes and molecular mechanisms associated with low egg production of broiler breeder hens in ad libitum

(Submitter supplied) Overfeeding reduces laying performance in broiler breeder hens (Gallus gallus domesticus). To unravel the effect of feeding regimes on energy metabolism and egg production, high-throughput RNA sequencing was utilized to identify differentially expressed genes (DEGs) in ovary, liver and adipose tissues of broiler chickens under ad libitum and restricted feeding. The transcriptome analysis showed that 289, 388 and 204 DEGs were identified in the adipose tissue, liver and ovary, respectively, between ad libitum and restricted feeding groups. more...

Long non-coding RNAs and mRNAs profiling of liver tissue during four developmental stages in chickens

(Submitter supplied) Liver is the major organ of lipid biosynthesis in chicken. In laying hens, the liver synthesizes most of yolk precursors and transports to the developing follicles to produce eggs. However, a systematic and comprehensive investigation of long non-coding RNAs (lncRNAs) and mRNAs transcriptome in chicken liver across different developmental stages remain clearly unknown. Here, we constructed 12 RNA libraries from liver tissue during four developmental stages, including juvenile period (JP, day 60), sexual maturity period (SM, day 133), peak laying period (PL, day 220) and broodiness period (BP, day 400). more...

Expression data from preadipocytes isolated from laying hen

(Submitter supplied) Adipogenesis is one of the most intensively studied models of cellular differentiation, and transcriptional activities involved in the process have been extensively investigated. We analyzed transcripts differentially expressed between hen preadipocytes treated with adipogenic cocktail containing 500 nM dexamethasone, 0.5 mM 3-isobutyl-1-methylxanthine, 20 ug/mL insulin and 300 uM OA (DMIOA) and non-treated control cells and transcripts differentially expressed between preadipocytes treated with DMIOA alone and those treated with a combination of DMIOA and 20(S)-hydroxycholesterol (DMIOA + 20(S)) using Affymetrix GeneChip Chicken Genome Array containing 28,000 transcripts.

Transcriptional Profiling of the Chicken Liver during the Embryo-to-Hatchling Metabolic Transition

(Submitter supplied) In the present study, we examined the hepatic transcriptome of chickens during the peri-hatch period—or the metabolic jump from chorioallantoic (embryo) to pulmonary (hatchling) respiration. Although our major interest was comparison of differentially-expressed genes between embryos and hatchlings, we made pairwise contrasts across six developmental ages. We collected the liver from four embryos at three ages (e16, e18 and e20) and four hatchling chicks at three ages (1, 3 and 9 days) post hatching. more...

Liver transcriptome analysis reveals extensive transcriptional plasticity during acclimation to low salinity in Cynoglossus semilaevis

(Submitter supplied) Background: Salinity is an important abiotic stress that influences the physiological and metabolic activity, reproduction, growth and development of marine fish. It has been suggested that half-smooth tongue sole (Cynoglossus semilaevis), a euryhaline fish species, use a large amount of energy to maintain osmotic pressure balance when exposed to fluctuations in salinity. To delineate the molecular response of C. more...
