Growth protocol |
Sprague-Dawley female rats were maintaned under a temperature-controlled environment and 12:12-h light:dark cycle. Adult females were fed either a control chow diet or a high-fat diet (42/45% energy from fat) for 12 weeks. Gonadal white adipose tissue was dissect and store at -80C until use.

Extraction protocol |
Total RNA was extracted using the RNeasy Lipid Tissue kit from Qiagen according to manufacturer’s instructions. RNA quality was confirmed using the Agilent 2100 Bioanalyzer (Agilent Technologies), and RNA concentration was measured on a NanoDrop (Thermo Scientific).

200 ng of total RNA was used as input to generate sense-strand cDNA with Ambion® WT Expression kit (Life Technologies: PN 4425209). The generated sense-strand cDNA was fragmented and biotin labelled using Affymetrix® GeneChip WT Terminal Labeling Kit (Affymetrix: PN 900671).
