Status Public on Oct 16, 2015
Title Asthmatic 9
Sample type RNA

Source name induced sputum
Organism Homo sapiens
Characteristics disease state: asthmatic
gender: male
age: 42
bmi: 28.5
smoking pack-years: none
ID: FAT079
Extracted molecule total RNA
Extraction protocol RNA was extracted from induced sputum using the RNeasy Mini Kit (Qiagen, Hilden, Germany);RNA quatitation was performed using the Quanti-iT RiboGreen RNA
Label biotin
Label protocol A total of 500 ng of sputum RNA was amplified and labelled with biotin-uridine triphosphate using the Total Prep RNA amplification kit as per manufacturer’s instructions (Life Technologies)

Hybridization protocol 750 ng cRNA was hybridised to Illumina Sentrix HumanRef-8 V3 Expression BeadChips (Illumina, San Diego, CA, USA) using standard protocols
Scan protocol Each BeadChip measured the expression of 24,354 genes and was scanned using the Illumina Bead Station and captured using BeadScan 3.5.11 (Illumina)
Description postmeal
Data processing Sample and gene profiling results were included in the analysis if the sample was of suitable purity (OD 260/280 ratio between 1.7-2.1) and was successfully amplified (sufficient cRNA generated) and hybridized (95th percentile of fluorescence score>500) and the data passed quality control criteria in GeneSpring GX (correlation coefficients and priciple component analysis plots)
