Title |
eye disc of ey > wge; Su(var)3-9[1]/[2], biologic rep 3 |
Sample type |
RNA |
Source name |
eye disc of ey > wge; Su(var)3-9[1]/[2]
Organism |
Drosophila melanogaster |
Characteristics |
genotype/variation: ey-Gal4 UAS-wge; Su(var)3-9[1]/[2] age: wandering 3rd instar larvae tissue: eye-antennal imaginal disc
Treatment protocol |
Thirty pairs of eye-antennal imaginal discs were dissected in cold PBS, and put them into Trizol reagent (Thermo Fisher Scientific).
Growth protocol |
Flies for each genotype were raised on a standerd cornmeal medium at room temperture. Third instar wandering larvae were dissected.
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Biotinylated aRNA were prepared by in vitto transcription of two-round synthesized cDNA, according to the standard protocol of GeneChip 3' IVT Express Kit, from 100 ng total RNA.
Hybridization protocol |
Following fragmentation, 12 ug of aRNA were hybridized for 16 hr at 45˚C on GeneChip Drosophila Genome 2.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol |
GeneChips were scanned using GeneChip Scanner 3000 (Affymeteix).
Description |
Drosophila_sv3
Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 30.
