Treatment protocol For monolayer based EC and SMC differentiation, pluripotent stem cells were first cultured in E8 medium, after replating, E8 medium was changed to RPMI1640 medium with AATS supplement with or without insulin, and with BMP4 and CHIR99021. After 3 days of induction, cells were replated and cultured in RPMI1640 medium with AATS supplement and VEGF and FGF. On differentiation day 8, CD31+ and SM22a+ cells were sorted and subjected to RNA-seq analysis.
Growth protocol Undifferentiated hESCs was cultured in E8 medium without feeders.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using RNeasy Plus Mini Kit (Qiagen) and treated with Rnase-free DNase.
RNA-seq library was prepared following the Smart-seq2 protocol (Picelli, 2014)

Data processing Illumina Casava software was used for basecalling.
Sequenced reads were trimmed for adaptor sequences, and masked for low-complexity or low-quality sequences, then mapped to hg38 genome using RSEM.
Fragments Per Kilobase per Million mapped reads (FPKM) of Refseq genes were caculated.
