Title |
5 days after treatment, biological rep 2 |
Sample type |
RNA |
Source name |
hearts from zebrafish at 5 days after 5-weeks of PHZ treatment
Organism |
Danio rerio |
Characteristics |
treatment: at 5 days after 5-weeks of PHZ treatment tissue: Heart
Treatment protocol |
PHZ was added to the fish water of the treated group for 30 min every 3 days for 5 weeks. The 30 min incubation with PHZ was followed by a 30 min wash in fish water. PHZ was used at a concentration of 1.25 μg/mL the first week followed by 2.5 μg/mL for the rest of the protocol. The control group followed the same process without the addition of PHZ in the fish water.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA from 3 biological replicates each containing 3 ventricles were isolated per time point using the RNeasy® micro kit (Qiagen). Potential contaminating genomic DNA was digested by DNase I treatment (Qiagen).
Label |
biotin
Label protocol |
250 ng of total RNAs were reverse transcribed into cDNA, then transcribed into cRNAs and labeled into biotinylated cRNA using the GeneChip WT Reagent kit (Affymetrix) according to the manufacturer’s standard protocols
Hybridization protocol |
Arrays were hybridized, washed and stained using the Affymetrix GeneChip WT Terminal Labeling and Hybridization kit.
Scan protocol |
GeneChip Scanner 3000 according to the manufacturer’s standard procedure
Description |
Gene expression data from the heart of zebrafish at 5 days after 5-weeks of PHZ treatment
Data processing |
Microarray data were pre-processed by Oligo R package using the robust multi-chip analysis (RMA)
