Three weeks old Arabidopsis seedlings were treated with 10µM estradiole for 5 hourse and then shoots were carefully harvested.. The leaf no.11 of wt and ANAC059 and ANAC092 T-DNA insertion lines were carefully harvested.

| Growth protocol |
Seeds were surface sterilized and sown in half-strength MS medium supplemented with 1% sucrose and solidified with 0.7% phytoagar and appropriate antibiotic. After inbibition, the seeds were strafied at 4C° for 3 days. The seeds were germinated at 22 C° under a 16-h day (140µmol m2- s1-) / 8-h night regime. The three weeks old Arabidopsis seedlings were removed carefully and subjected to the treatment. T-DNA insertion lines were grown in soil and germinated under controlled condition in a growth chamber with a 16-h day lenght provided by fluorescent light at 80 or 120 µmol m2- s1- and a days/night temperature of 20/16 C° and relative humidity 60/75%.
