Treatment protocol |
Patients were treated with individually dosed rFSH (Puregon®, MSD, Denmark) or hMG (Menopur®, Ferring, Denmark) starting on day 2-3 of the menstrual cycle. From stimulation day 5-6 a GnRH antagonist (Ganirelix 0,25 mg (Fyremadel®, SUN pharma, Netherlands)) was administered daily. When at least three follicles reached ≥17 mm in diameter, an ovulation trigger (hCG (6500 IU (Ovitrelle®, Merck Serono, Germany) or GNRHa (buserelin 0,5 mg (Suprefact®, Sanofi-Aventis)) was administered and oocyte pick up was planned for 36 hours later.

Growth protocol |
Granulosa cells were isolated from aspirated follicular fluid with a pipette as blood clots and vessels were avoided. The cells were washed through a 4-well dish containing PBS and 0,1% PVA, transferred to a cryotube with minimal PBS, snap frozen in liquid nitrogen and stored at -80⁰C until RNA extraction.

RNA was processed using Clariom™ D Pico Assay (Applied Biosystems, Thermo Fisher, USA) according to the manufacturer’s protocol, using the Affymetrix Fluidics Station 450.

Scan protocol |
Arrays were scanned in the Affymetrix GeneArray 3000 7G scanner to generate fluorescent images. Cell intensity files (.CEL files) were generated in the GeneChip Command Console Software (AGCC, Affymetrix, Thermo Fisher, USA).

Data processing |
The 83 raw CEL-files were imported into Transcriptome Analysis Console (TAC 4.0.1, applied Biosystems, Thermo Fisher Scientific, MA, USA) . Normalization and summarization were performed by the SST-RMA (Robust Multichip Analysis) approach.
