Growth protocol protonemata were  cultivated axenically in petri dishes  containing PpNH4 medium supplemented with 7g.L-1 agar in a growth chamber maintained at 37 °C under a photoperiod of 16/8 h and an irradiance of 100 µmol.m-2.s-1. Harvest at 12H.
Extracted molecule total RNA
Extraction protocol Total moss RNA was extracted from 100 mg of P. patens 7-day protonemal tissues from control or heat-treated samples using Nucleospin® RNA plant (Macherey-Nagel-Germany).
Sequencing was carried out by Eurofin (Germany) using one channel and Illumina Illumina HiSeq 2500 2x 100 bp paired-end. RNA libraries were prepared for sequencing using standard Illumina protocols
strand-specific cDNA libraries; RNA selection using polyA selection. RNAseq
