Treatment protocol Cells were incubated with 100 and 1000 nM talazoparib (MedChemExpress, #HY-16106) or 0.005% DMSO (Sigma, #D5879) for 48h. After the drug incubation, cells were washed twice with 1xPBS (Gibco, #20010019) and collected after trypsinization.
Growth protocol 250,000 - 400,000 cells were seeded in 60 mm cell culture dishes (Falcon, #353002) in DMEM/F12 growth media (Thermo Fischer Scientific #31330038) supplemented with 15% Fetal Bovine Serum (Thermo Fischer Scientific #10270106) and 100 U/ml penicillin-streptomycin (Thermo Fischer #15140122). The cells were recovered for 24h before treatment.
Extracted molecule total RNA
Extraction protocol Total RNA was isolated from cell pellets using the Qiagen Allprep DNA/RNA/miRNA Universal kit, according to the manufacturer's protocol (Qiagen).
Label biotin
Label protocol RNA was amplified, fragmented and labelled according to the Affymetrix GeneChip HT WT User Guide.
 
Hybridization protocol Samples were hybridized onto Affymetrix Human Transcriptome 2.0 Arrays according to the manufacturer's protocol (Affymetrix).
Scan protocol Array scanning was performed according to the manufacturer's protocol (Affymetrix).
Description SST RMA expression values at the gene-level derived using Expression Console software
Data processing Gene-level expression signals were obtained from raw data CEL files by the SST RMA procedure implemented in the Affymetrix Expression Console software.
