Treatment protocol |
4 weeks post-MI, mice were sacrificed by isofluorane and cervical dislocation. Hearts were explanted and the peri-infarct region was identified via microscopy, dissected out. Samples were rinsed with PBS twice and placed in Trizol (Thermo Fisher Scientific).

Growth protocol |
Female 80-100 day-old SCID mice were purchased from Charles River Laboratories (MA). iCMs were differentiated from iPSCs and cultured until spontaneous contractions were observed. iCM-Ex were precipitated from iCMs.

Extraction protocol |
Trizol extraction of total RNA was performed using miRNeasy Mini Kit (Qiagen) according to the manufacturers’ instruction.

5.5 μg of sense-strand cDNA was labeled with biotin according to the Clariom S microarray protocol (Thermo Fisher Scientific).

Hybridization protocol |
Fragmented and labeled ss-cDNA were hybridized for 16 hr at 45°C on rotation onto Clariom S Mouse Array gene chips (Thermo Fisher Scientific).

Scan protocol |
Arrays were scanned using Affymetrix GeneChip Scanner.

Data processing |
Data were analyzed with Expression Console and Transcription Analysis Console softwares (Thermo Fisher Scientific) using default settings.
