Extraction protocol |
Tumor material was subjected to pathological frozen section; and immediately snaps frozen. 0.5 cm3 of tissue was lysed using Lysing Matrix A (Q-BIOgene) and were homogenized (BIO101/Savant FastPrep Homogenizer). After an overnight incubation with SDS and proteinkinase K, DNA was precipitated by adding ice-cold isopropanol, collected and dried. Concentration and quality of dissolved DNA was measured using Nanodrop Spectrophotometer (Parmacia Biotech).
