20 male and 20 female Drosophila brain of the appropriate geneotype were dissected were dissected on ice cold Schneider’s medium. After a brief wash with PBS, the brains were incubated with 0.05% trypsin-EDTA (Fisher scientific) for 30 minutes, with continuous pipetting every 5 minutes. The dissociated tissue were filtered through a 10um cell strainer to get a single cell suspension. 10x libraries were constructed according to the manufacter’s instructions (Chromium Next GEM Single Cell 3' Kit v3.1).

The demultiplexing, barcoded processing, gene counting and aggregation were made using Cellranger, version 6.0, 10x Genomics.(https://support.10xgenomics.com/single-cell-gene-expression/software/pipelines/latest/what-is-cell-ranger) For Control 1, Control 3 and tauP251L KI 3 libraries were sequenced once with enough sequencing depth. For control 2, tau P251L KI 1 and tau P251L KI 2, the libraries were sequenced twice to get enough sequencing depth.
