Sample GSM7220533 Query DataSets for GSM7220533
Status Public on Oct 03, 2023
Title Anesthesia with sevoflurane
Sample type SRA

Source name liver
Organism Rattus norvegicus
Characteristics tissue: liver
genotype: wild type
treatment: Subjection of anesthesis with sevoflurane for 6h
time: 6 hours
Treatment protocol Rats were subjected to inhalational anesthesia using either sevoflurane (4.5%) or desflurane (6%) and rats were sacrificed by decapitation immediately after their loss of consciousness or after subjection of inhalational anesthesia for 6 hours and then portions of their livers were recovered and immersed in RNAlater until RNA preparation.
Extracted molecule total RNA
Extraction protocol Total RNAs were harvested using RNeasy Midi Kit (Qiagen).
RNA libraries for RNA-seq were prepared using TruSeq Stranded Total RNA Library Prep Gold Kit (illumina) according to manufacturer’s instruction.

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina NovaSeq 6000

Data processing Terminal
Sequence reads were trimmed for adaptor sequence/low-quality sequence using sickle (parameter- -q 30 -l 20 )
Trimmed sequence reads were mapped to rn6 using hisat2 and samtools
Read count extraction and normalization were performed using Stringtie
Assembly: rn6
Supplementary files format and content: tab-delimited text files include RPKM and TPM values for each Sample
