Treatment protocol To obtain MDSC subpopulations, CD11b+ cells, enriched by the MACS system, were stained with APC-conjugated anti-CD11b Ab, FITC-conjugated anti-Ly-6C Ab and PE-conjugated anti-Ly-6G Ab and sorted using a FACSAria II (BD Bioscience).
Growth protocol BALB/c mice were subcutaneously injected with 1x10^5 Her-2/CT26 cells.
Extracted molecule total RNA
Extraction protocol Total RNA was purified from CD11b+Ly-6C(high)Ly-6G(low) and CD11b+Ly-6C(low)Ly-6G(high) cells using Qiagen RNeasy kits (Qiagen, Valencia, CA).
Label Biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 100 ng total RNA (Wt_sensetarget_label_manual(updated manual)_1, 2007, Affymetrix).
 
Hybridization protocol Hybridization cocktails containing 5 to 5.5 µg of fragmented, end-labeled single-stranded cDNA were prepared and hybridized to GeneChip Human Gene 1.0 ST arrays for 17hr at 45°C. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the GeneChip Scanner 3000 7G.

Data processing The RMA algorithm was used for expression summary and signal calculation of Affymetrix MoGene 1.0 ST array data. Sketch Quantile normalization was performed for the signal normalization.
