Transcriptional Profiling of Hypoxic Neural Stem Cells to Identify Novel Regulators of Neural Stem Cell Biology

(Submitter supplied) Neural stem cells reside in a hypoxic microenvironment within the brain. However, the crucial transcription factors that regulate neural stem cell biology under physiologic hypoxia are poorly understood. Here, we have performed microarray analysis of hypoxic versus normoxic neural stem cells with the aim of identifying pathways and transcription factors that are activated under oxygen concentrations mimicking normal brain tissue microenvironment.

Unbiased transcriptome profiling of mNS-5 (mouse embryonic stem cell-derived neural stem cells) following RNAi-mediated MED12 depletion.

(Submitter supplied) The RNA polymerase II transcriptional Mediator subunit MED12 is broadly implicated in vertebrate brain development, and genetic variation in human MED12 is associated with X-linked intellectual disability and neuropsychiatric disorders. Although prior studies have begun to elaborate the functional contribution of MED12 within key neurodevelopmental pathways, a more complete description of MED12 function in the developing nervous system, including the specific biological networks and cellular processes under its regulatory influence, remains to be established. more...

Identification of Tox chromatin binding properties and of its downstream targets by DamID

(Submitter supplied) Purpose: The goal of this study was to characterize the chromatin binding features and the transcriptional target genes of a new key player during corticogenesis, the transcription factor Tox Methods: DamID was performed using two cell lines, the Neuro-2a and the HEK-293T cells. Experiments were performed in duplicates for each cell line by infecting them with pLgwV5Eco-ToxDam lentiviruses and using the pLgxV5Dam viruses as control. more...

Cerebellar Granule Neuron Temporal-NFI Microarray Data

(Submitter supplied) Dendrite and synapse development are critical for establishing appropriate neuronal circuits, and disrupted timing of these events can alter connectivity leading to disordered neural function. In early postnatal development of cerebellar granule neurons (CGNs), the expression of many genes is temporally regulated. Further, NFI (Nuclear Factor I) proteins have been shown to play important roles in the regulation of gene expression in developing CGNs. more...

Single-Cell Transcriptomics Reveals a Population of Dormant Neural Stem Cells that Become Activated upon Brain Injury

(Submitter supplied) Heterogeneous pools of adult neural stem cells (NSCs) contribute to brain maintenance and regeneration after injury. The balance of NSC activation and quiescence, as well as the induction of lineage-specific transcription factors, may contribute to diversity of neuronal and glial fates. To identify molecular hallmarks governing these characteristics, we performed single-cell sequencing of an unbiased pool of adult subventricular zone NSCs. more...
