Treatment protocol Medium supplemented with 0.4% DMSO was changed every 2-3 days. For PHH spheroids cells were also treated with either aflatoxin B1 (0.002uM), amiodarone (4uM) or chlorpromazine (6uM).
Growth protocol Primary human hepatocytes were seeded in ultra-low attachment plates at 1500 cells/well in 100ul Williams E basal medium supplemented with 10% FBS and centrifuged at 100g for 2 minutes. From day 4 onwards daily medium changes with FBS-free medium were performed. Treatments were initiated on day 7.
Extracted molecule total RNA
Extraction protocol RNA was extracted with the AllPrep DNA/RNA Mini Kit (Qiagen) according to the manufacturer’s instructions. RNA concentration and quality was assessed using the Agilent Bioanalyzer
Label Cy3
Label protocol Total RNA samples (45ng) were labelled with Cy3 using the Low Input Quick Amp Labeling Kit–One Colour (Agilent# 5190-2305)
 
Hybridization protocol Agilent 8x60K Whole Human Genome Oligo Microarray slides (G4851B) were hybridised, washed and then scanned on an Agilent Microarray Scanner
Scan protocol Agilent 8x60K Whole Human Genome Oligo Microarray slides (G4851B) were hybridised, washed and then scanned on an Agilent Microarray Scanner

Data processing Images from the Agilent scanner were processed using Agilent Feature Extraction Software v10.7.3.1.
