Identification of COX-2 as a major actor of transcriptomic adaptation of endothelial and tumor cells to cyclic hypoxia

(Submitter supplied) Tumor hypoxia is not a stable phenomenon but cycles between periods of deep hypoxia and reoxygenation. Cyclic hypoxia originates from heterogeneities in red blood cell flux and from the permanent remodelling of the angiogenic vascular network. Endothelial cells lining tumor blood vessels are therefore also influenced by cyclic hypoxia. The gene expression pattern promoted by cyclic hypoxia differs from those observed under normoxia and even continuous hypoxia. more...

Transcriptome analysis of pancreatic cancer cell line that differ in metastatic potential

(Submitter supplied) Two pancreatic cancer cell lines with different metastatic and growth potential were compared under hypoxic conditions and under normal atmospheric oxygen pressure. The FG cell lines shows very few metastases and slow growth in mouse xenograft models. L3.6pl, derived from FG by cycles re-implantation of metastatic cells obtained after orthotopic tumor growth in nude mice, shows high motility, aggressive growth and very high metastatic potential By comparison of the two cell lines under different oxygen concentration we tried to simulate in vivo conditions of tumors at different growth stages. more...

Identification of PDGF-dependent patterns of gene expression in U87 glioblastoma cells

(Submitter supplied) Overall study: Identification of PDGF-dependent patterns of gene expression in U87 glioblastoma cells. RNA was obtained from triplicate dishes of 5 different groups of U87 cells, each (total 15) analyzed with one U95 microarray chip. Three different comparisons were made: 1) Clone 3.1 (34580-34582) vs. clone 3.3 (34583-34585) vs. parent U87 (34592-34594). Purpose: demonstrate that the gene expression profiles between these 3 cell lines are not different, so they could be pooled as a single untreated group. more...

Testicular germ cell tumors and their histological subgroups

(Submitter supplied) Normal, premalignant and various histological subtypes of testicular germ cell tumor (TGCT) tissues were hybridized against Universal Human Reference RNA (Stratagene) onto Agilent 60mer oligo microarrays (GEO accession no GPL885). In vitro time series of two TGCT cell lines, NTERA2 and 2102Ep, treated with retinoic acid for 0, 3, and 7 days were also included. The data set (30 hybridizations) is particularly useful for comparisons between various histological subtypes of TGCT versus each other or versus normal testis. more...

Human Glioblastoma

(Submitter supplied) Summary: Astrocytomas can be categorized as either low grade or high grade (glioblastoma). Low grade astrocytomas are not generally aggressive tumors whereas glioblastomas are and in turn have a high mortality rate. The purpose of this experiment is to identify genetic differences between the two types. Hypothesis: There will be a difference in RNA expression between high grade and low grade tumors. more...

Comparative genome analysis of exosomal mRNA derived from Tspan8-overexpressing and Tspan8-negative rat pancreas tumors

(Submitter supplied) We recently described that tumor-derived exosomes containing the tetraspanin Tspan8 induce angiogenesis. However, not much is known about exosome-endothelial cell interactions and there are no clues on the role of tetraspanins, which are a constitutive component of exosomes. We used the stably Tspan8 expressing rat adenocarcinoma cell line AS-Tspan8, to explore how exosomal Tspan8 supports angiogenesis. more...

Hypoxic response in wild type and HIF-1alpha null hepatoctyes

(Submitter supplied) The primary aim of this study was to evaluate the changes in hepatocyte gene expression under short-term hypoxic conditions in wild type and HIF-1a null cultures. To this end, hypoxia treated cultures were subjected to incubation with 1% O2/5% CO2/94% N2 at 37° C for eight hours prior to RNA isolation. Duplicate normoxic controls were established from separate animals wherein cultures were untreated and treated with Adbgal. more...

Expression data from TAp73+/+ and TAp73-/- MEFs

(Submitter supplied) The aim of the experiment was to analyse the gene expression differences between MEFs containing TAp73 or without TAp73, at basal levels in normal culture conditions, and upon treatment with hypoxia (by incubation in 1% oxygen) for 4 or 8 hrs. The analysis indicate that absence of TAp73 leads to changes in multiple cellular and biological processes.

A novel tumor-promoting function resided in the 5’ untranslated region of vegf mRNA

(Submitter supplied) VEGF165 is one of the key regulators of tumor development. Using HCT116 cells transfected with full-length vegf mRNA, full-length vegf mRNA with mutations of H9D and L14E, mutated vegf mRNAs lacking untranslated regions (UTRs), and 5’UTR mutated between nt 591 and nt 746, we found that vegf 5’UTR resided an anti-apoptotic activity against chemotherapy independently of VEGF165. We next established HCT116 clones stably expressing vegf 5’UTR or the mutated vegf 5’UTR. more...

Iron Chelators Treatment on MCF-7 Human Breast Cancer Cell

(Submitter supplied) Iron-deficiency affects 500 million people, yet the molecular role of iron in gene expression remains poorly characterized. Moreover, the alterations in global gene expression after iron chelation remains unclear and are important to assess for understanding the molecular pathology of iron-deficiency and the biological effects of iron chelators. We assessed the effect on whole genome gene expression of two iron chelators (desferrioxamine and 2-hydroxy-1-napthylaldehyde isonicotinoyl hydrazone) that have markedly different permeability properties. more...

Expression analysis of LM8 osteosarcoma cells overexpressing human Prolyl Hydroxylase Domain Protein-4 (PHD4) and LM8 control cells

(Submitter supplied) PHD4 regulates the expression of Hypxia-inducible Factor 2 (HIF-2) alpha in LM8 osteosarcoma cells. PHD4 overexpression inhibits the growth of experimental tumor in syngenic mice but stimulates angiogenesis via Transforming Growth-Factor (TGF)-alpha. We used microarrays to investigate the gene expression of stable LM8 cell line overexpressing human PHD4 in comparison to the LM8 control line, transfected with the empty vector.
