Trans-10, cis-12 conjugated linoleic acid delipidiative effect on white adipose tissues: time course

Analysis of retroperitoneal white adipose tissue (WAT) of C57BL/6J males fed a diet containing 0.5% trans-10 ,cis-12 conjugated linoleic acid (t10c12 CLA) for up to 17 days. Results provide insight into the mechanisms underlying the reduction in fat content of mice WAT in response to t10c12 CLA.

Microarray Analysis of the Delipidation of White Adipose Tissue of Mice Fed Conjugated Linoleic Acid

(Submitter supplied) The white adipose tissue (WAT) rapidly loses mass when mice are fed a diet containing trans-10, cis-12 conjugated linoleic acid (t10c12 CLA). A microarray analysis of WAT due to CLA feeding was performed to better define the processes and genes involved. WAT weight decreased by ca. 80% over 17 days of feeding a 0.5% t10c12 CLA diet. Trans-10, cis-12 conjugated linoleic acid (t10c12 CLA) reduces triglyceride levels in adipocytes. Conjugated linoleic acid activates AMPK and reduces adiposity more effectively when used with metformin

(Submitter supplied) Trans-10, cis-12 conjugated linoleic acid (t10c12 CLA) causes dramatic reductions in white adipose tissue in mice but has had limited effectiveness in humans. Determination of the signaling pathways involved may lead to better regulation of adiposity. T10c12 CLA was found to activate AMP-activating protein kinase (AMPK), a central regulator of cell metabolism. Gene expression profile of peroxisome proliferator-activated receptor delta (Ppard)-overexpressing RAW 264.7 macrophages treated vehicle, GW501516 or interleukin-4 (Il-4)

(Submitter supplied) Investigation of whole genome gene expression level changes in GW501516 (a Ppard ligand) or Il-4 treated Ppard-overexpressing RAW 264.7 macrophages as compared to vehicle. Alternative activation of adipose tissue resident macrophages inhibits obesity-induced metabolic inflammation. In the current study we profile genes regulated by two M2 inducers, Il-4 or Ppard agonists and find that alternative activation promotes the cell survival program, while inhibiting that of inflammation-related cell death.
